Fängerantikörper 1.0 mg/mL of rabbit anti-mouse PIGR monoclonal antibody, Dilute to a working concentration of 2.0 μg/mL in CBS before coating.

Nachweisantikörper 0.5 mg/mL rabbit anti-mouse PIGR polyclonal antibody conjugated to horseradish-peroxidase (HRP). Dilute to working concentration of 0.25 μg/mL in detection antibody diluteion buffer before use.

Standard Each vial contains 50 ng of recombinant mouse PIGR. Reconstitute standard powder with detection antibody dilution buffer. After reconstitution, store at -20℃ to -70℃ in a manual defrost freezer. A seven-point standard curve usi ng 2-fold serial dilutions in sample dilution buffer, and a high standard of 500 pg/mL is recommended.

The minimum detectable dose of Mouse PIGR was determined to be approximately 7.81 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.

The Sino Biological ELISA Pair Set is a solid phase sandwich ELISA (Enzyme-Linked Immunosorbent Assay). It utilizes a monoclonal antibody specific for PIGR coated on a 96-well plate. Standards and samples are added to the wells, and any PIGR present binds to the immobilized antibody. The wells are washed and a horseradish peroxidase conjugated rabbit anti-PIGR polyclonal antibody is then added, producing an antibody-antigen-antibody "sandwich". The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount of PIGR present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.

Polymeric immunoglobulin receptor, also known as PIGR, is a member of the immunoglobulin superfamily and a Fc receptor. The ectodomain of this receptor consists of five units with homology to the variable units of immunoglobulins and a transmembrane region, which also has some homology to certain immunoglobulin variable regions. PIGR is expressed on several glandular epithelia including those of liver and breast. The deduced amino-acid sequence has a length of 764 residues and shows an overall similarity of 56% and 64% with the rabbit and rat counterpart. PIGR mediates transcellular transport of polymeric immunoglobulin molecules, and thus facilitates the secretion of IgA and IgM. During this process, a cleavage occurs that separates the extracellular (known as the secretory component) from the transmembrane segment of PIGR.
