Fängerantikörer 0.15 mg/mL of mouse anti-DLL4 monoclonal antibody, Dilute to a working concentration of 2.0 μg/mL in CBS before coating.

Nachweisantikörper 0.5 mg/mL rabbit anti-DLL4 monoclonal antibody conjugated to horseradish-peroxidase (HRP). Dilute to working concentration of 0.25 μg/mL in detection antibody diluteion buffer before use.

Standard Each vial contains 70 ng of recombinant DLL4. Reconstitute standard powder with detection antibody dilution buffer. After reconstitution, store at -20℃ to -70℃ in a manual defrost freezer. A seven-point standard curve usi ng 2-fold serial dilutions in sample dilution buffer, and a high standard of 2000 pg/mL is recommended.

2. Sensitivität

The minimum detectable dose of Human DLL4 was determined to be approximately 31.25 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.

Standards and samples are added to the wells, and any DLL4 present binds to the immobilized antibody. The wells are washed and a horseradish peroxidase conjugated rabbit anti-DLL4 monoclonal antibody is then added, producing an antibody-antigen-antibody "sandwich". The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount of DLL4 present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.

Speicherung - Detection Antibody should be protected from prolonged exposure to light. Aliquot the reagents and store at -20℃ to -70℃ in a manual defrost freezer.

Delta-like protein 4 ( DLL4 ) is a type I membrane-bound Notch ligand characterized by an extracellular region containing several EGF-like domains and a DSL domain required for receptor binding, and is suggested to play a key role in vascular development and tumor angiogenesis. The Notch pathway is an evolutionary conserved intercellular signaling pathway involved in numerous biological processes including cell fate determination, cellular differentiation, proliferation, survival, and apoptosis. In mammalian cells, five Notch ligands (Jagged1, 2, DLL1, 3, 4) and four Notch receptors (Notch1-4) have been identified, and ligand-receptor interactions results in proteolysis and translocation of the Notch intracellular domain. As a ligand for Notch1 and Notch4, DLL4 is selectively expressed in the developing endothelium and in some tumor endothelium, and is induced by vascular endothelial growth factor (VEGF)-A and hypoxia. In recent studies, it has been revealed that this DLL4 inhibition may paradoxically lead to increased angiogenesis but poor tumor growth because of the non-functional. neovascularization, and constitutive expression of DLL4 may leads to a lethal lymphoproliferative disease. Accordingly, this may provide a new therapeutic approach for certain carcinomas.
