PERP1, also known as MZB1, is an endoplasmic reticulum-localized and B cell-specific protein that is most abundantly expressed in marginal zone (MZ) B and B1 cells. PERP1 helps to diversify peripheral B cell functions by regulating Ca(2+) stores, antibody secretion, and integrin activation. PERP1 also associates with immunoglobulin M (IgM) heavy and light chains and promotes IgM assembly and secretion. It may exert its effect by acting as a molecular chaperone or as an oxidoreductase as it displays a low level of oxidoreductase activity. Methylation-mediated silencing of MZB1 expression leads to loss of its tumor-suppressive activity, which may be a factor in the hepatocarcinogenesis, and is a useful prognosticator in HCC.
