Interleukin-5 receptor subunit alpha, also known as IL-5 receptor subunit alpha, IL-5R subunit alpha, IL-5R-alpha, IL-5RA and CD125, is a single-pass type I membrane protein which belongs to the type I cytokine receptor family and type 5 subfamily. IL5Ra / CD125 is expressed on eosinophils and basophils. IL5Ra / CD125 is the receptor for interleukin-5. The alpha chain binds to IL5. It is a heterodimer of an alpha and a beta subunit. The beta subunit is common to the IL3, IL5 and GM-CSF receptors. It interacts with SDCBP. The WSXWS motif of IL5Ra / CD125 appears to be necessary for proper protein folding and thereby efficient intracellular transport and cell-surface receptor binding. The box 1 motif of IL5Ra / CD125 is required for JAK interaction and/or activation. The binding of this protein to IL5 depends on the beta subunit. The beta subunit is activated by the ligand binding, and is required for the biological activities of IL5. This protein has been found to interact with syndecan binding protein (syntenin), which is required for IL5 mediated activation of the transcription factor SOX4.
