Contactin-5 (CNTN5), also known as NB-2, is an membrane-anchored neuronal protein and also an adhesion molecule. CNTN5 is one of the neural recognition molecules in the contactin subgroup, which belongs to the immunoglobulin superfamily. CNTN5 shared 46, 43, 55 and 55% identities with CNTN1,CNTN2,CNTN3,CNTN4 respectively at the amino acid sequence level. Expression of NB-2 mRNA was restricted to cerebrum, cerebellum and was hardlty detectable, if any, in spinal cord. In humans, two isoforms have been identified. The long isoform of human NB-2 consists of 1100 amino acids residues and the short isoform lacks 74 amino acid residues between residues 19 and 93 of the long isoform. CNTN5 may play a role in the developing nervous system. NB-2 promoted neurite outgrowth of the cerebral cortical neurons but not the hippocampal neurons. CNTN5-deficient mice demonstrate impaired neuronal activity in the auditory system, suggesting that it plays specific roles in the development and/or maturation of the regions.
