Hsp90 co-chaperone CDC37, also known as Hsp90 chaperone protein kinase-targeting subunit, cell division cycle 37 homolog, CDC37 and CDC37A, is a cytoplasm protein which belongs to the CDC37 family. CDC37 is monomeric in its physiological state, and that it becomes a dimer only when it is complexed with both Hsp90 and protein kinases. CDC37 is a co-chaperone that binds to numerous kinases and promotes their interaction with the Hsp90 complex, resulting in stabilization and promotion of their activity. CDC37 protein specifically interacts with Cdk4 and Cdk6, but not with Cdc2, Cdk2, Cdk3, Cdk5 and any of a number of cyclins tested. CDC37 is not an inhibitor nor an activator of the Cdk4 / cyclin-D1 kinase, while it appears to facilitate complex assembly between Cdk4, and cyclin-D1. CDC37 competes with p16 for binding to Cdk4, suggesting that p16 might exert part of its inhibitory function by affecting the formation of Cdk4/cyclin D1 complexes via CDC37. CDC37 is a cell division cycle control protein of Saccharomyces cerevisiae. It is a molecular chaperone with specific function in cell signal transduction. It has been shown to form complex with Hsp90 and a variety of protein kinases including CDK4, CDK6, SRC, RAF1, MOK, as well as eIF-2 alpha kinases. CDC37 is thought to play a critical role in directing Hsp90 to its target kinases.
