1. Materialien gebietet

Fängerantikörper 0.5 mg/mL of mouse anti-CD147 monoclonal antibody, Dilute to a working concentration of 4.0 μg/mL in CBS before coating.

Nachweisantikörper 0.5 mg/mL rabbit anti-CD147 monoclonal antibody conjugated to horseradish-peroxidase (HRP). Dilute to working concentration of 0.125 μg/mL in detection antibody diluteion buffer before use.

Standard Each vial contains 25 ng of recombinant CD147. Reconstitute standard powder with detection antibody dilution buffer. After reconstitution, store at -20℃ to -70℃ in a manual defrost freezer. A seven-point standard curve usi ng 2-fold serial dilutions in sample dilution buffer, and a high standard of 400 pg/mL is recommended.

2. Sensitivität

The minimum detectable dose of Human CD147 / EMMPRIN was determined to be approximately 6.25 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.

The wells are washed and a horseradish peroxidase conjugated rabbit anti-CD147 / EMMPRIN monoclonal antibody is then added, producing an antibody-antigen-antibody "sandwich". The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount of CD147 / EMMPRIN present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.

STORAGE - Detection Antibody should be protected from prolonged exposure to light. Aliquot the reagents and store at -20℃ to -70℃ in a manual defrost freezer.

CD147, also known as Basigin, BSG, and EMMPRIN, is a transmembrane glycoprotein with different forms resulted from different modes of glycosylation and N-terminal sequence variants. CD147 is a member of the immunoglobulin superfamily with homology to both the immunoglobulin V domain and MHC class II antigen beta-chain. This protein play important roles in variety of events including spermatogenesis, embryo implantation, neural network formation, and tumor progression. Enriched on the plasma memebrane of many human cancer (breast cancer, lymphoma, oral squamous cell carcinoma, glioma, melanoma, lung, bladder and kidney carcinomas ) cells, CD147 induces the production and release of matrix metalloproteinases in the surrounding mesenchymal cells and tumor cells, and thereby promotes invasion, metastasis, growth and survival of malignant cells. Furthermore, CD147 also serves as a receptor for extracellular cyclophilinthe and its association with integrins might be important in signal transduction.
