B7-H1, also known as CD274 and PD-L1, is a cell surface glycoprotein belonging to the B7 family of costimulatory molecules. B7-H1 is originally identified by homology analysis in comparison with B7-1 and B7-2, two molecules with important immunoregulatory functions. B7-H1 costimulates T-cell proliferation and preferentially induces IL-10 and IFN-gamma secretion in a PDCD1-independent manner. The costimulatory function of B7-H1 is critical for enhancing maturation and differentiation of T-cells in lymphoid organs. On contrast, B7-H1 also functions as a co-inhibitor through programmed death-1 (PD-1). By binding to PD-1 receptors on activated T-cells and B-cells, B7-H1 inhibits T-cell proliferation by blocking cell cycle progression and cytokine production, and thus inhibits ongoing T-cell responses in peripheral tissues. Aberrantly expressed of B7-H1 by tumor cells or lymphocytes is associated with aggressive pathologic features of certain carcinomas and represents a promising target for immune manipulation. Mouse and human B7-H1/PD-L1 share 73% amino acid sequence identity.
