Apoptosis is an essential processes required for normal development and homeostasis of all metazoan organisms. Apoptosis is carried out by caspases which reside in cells as inactive zymogens, and the initiator caspase zymogens are activated by adaptor proteins such as FADD and Apaf-1 mediated either by death receptors or cytochrome c. DIABLO (direct IAP binding protein with low pI), also well known as SMAC (second mitochondria derived activator of caspase), is a mitochondrial homodimeric protein, and performs a critical function in apoptosis by eliminating the inhibitory effect of IAPs (inhibitor of apoptosis proteins) on caspases. It has been shown that DIABLO mainly interacts with IAPs in the cytochrome c/Apaf-1/caspase-9 pathway, and promotes apoptosis. DIABLO is released from the mitochondria into the cytosol occurring downstream of cytochrome c release in response to apoptotic stimuli such as irradiation, DNA damage or cytotoxic drugs.
