VDR(vitamin D(1,25- dihydroxyvitamin D3)receptor, Histidine-tagged) , also known as NR1I1, belongs to the NR1I family, NR1 subfamily. It is comprised by three domains: a modulating N-terminal domain, a DNA-binding domain and a C-terminal ligand-binding domain. VDR can be detected in the intestine, thyroid and kidney and has a vital role in calcium homeostasis. VDR functions as nuclear hormone receptor and mediates the action of vitamin D3 by controlling the expression of hormone sensitive genes. It regulates transcription of hormone sensitive genes via its association with the WINAC complex, a chromatin-remodeling complex. VDR also plays a central role in calcium homeostasis and functions as a receptor for the secondary bile acid lithocholic acid. Defects in VDR are the cause of rickets vitamin D-dependent type 2A. A disorder of vitamin D metabolism can cause severe rickets, hypocalcemia and secondary hyperparathyroidism.
