Neurexophilin-1 ( NXPH1 ) is a member of the neurexophilin family, this secreted protein with a variable N-terminal domain, a highly conserved, N-glycosylated central domain, a short linker region, and a cysteine-rich C-terminal domain. Neurexophilin-1 protein is encoded by the NXPH1 gene, which has highest level in brain. It was discovered as a neuronal glycoprotein that is copurified with neurexin Ialpha during affinity chromatography on immobilized alpha-latrotoxin. Neurexophilins represent a family of extracellular signaling molecules that interact with multiple receptors including all three alpha-neurexins. The mammalian brains contain four genes for neurexophilins the products of which share a common structure composed of five domains. Neurexophilins 1 and 3 but not 4 bind to a single individual LNS domain, the second overall LNS domain in all three alpha-neurexins. Neurexophilin-1 may be proteolytically processed at the boundary between the N-terminal non-conserved and the central conserved domain in neuron-like cells. As a secreted cysteine-rich glycoprotein, Neurexophilin-1 is synthesized in a subclass of neurons, and may be signaling molecules that resemble neuropeptides.
