This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Human IDH1 (rh IDH1; Catalog#12055-H08E; O75874; Met 1-Leu 414).

Flow cytometric analysis of Human IDH1 expression in HepG2 cells. The cells were treated according to manufacturer's manual (BD Pharmingen™ Cat. No. 554714), and stained with Purified Mouse anti-IDH1 (12055-MM03), then a FITC-conjugated second step antibody. The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.

Flow cytometry was performed on a BD FACSCalibur flow cytometry system

Storage This antibody can be stored at 2℃-8℃ for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -70℃. Preservative-Free.
Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

Hintergrund von IDH1 Antikörper

Isocitrate dehydrogenases 1 (IDH1), one of the Isocitrate dehydrogenases, which catalyze the oxidative decarboxylation of isocitrate to α-ketoglutarate and reduce NAD (P)+ to NAD (P)H. This process involves oxidation of isocitrate to oxalosuccinate, with NAD (P)H as the electron acceptor, followed by decarboxylation of oxalosuccinate to form α-ketoglutarate. IDH1 and IDH2 function at a crossroads of cellular metabolism in lipid synthesis, cellular defense against oxidative stress, oxidative respiration, and oxygen-sensing signal transduction. IDH1 and IDH2 mutations are usually heterozygous in cancer, and they appear to confer a neomorphic enzyme activity for the enzymes to catalyze the production of D-2-hydroxyglutarate. IDH1 is highly expressed in the mammalian liver and moderately expressed in other tissues. It contains a C-terminal tripeptide peroxisome targeting signal 1 sequence. Isocitrate dehydrogenase 1 (IDH1) mutations have recently been identified as early and frequent genetic alterations in astrocytomas, oligodendrogliomas, and oligoastrocytomas, as well as secondary glioblastomas, whereas primary glioblastomas very rarely contain IDH1 mutations.
