CD63 antigen, also known as Granulophysin, Lysosomal-associated membrane protein 3, Melanoma-associated antigen ME491, Ocular melanoma-associated antigen, Tetraspanin-30, CD63 and Tspan-30, is a multi-pass membrane protein which belongs to thetetraspanin (TM4SF) family. Tetraspanins comprise a large superfamily of cell surface-associated membrane proteins characterized by four transmembrane domains. Tetraspanins participate in a variety of cellular processes, like cell activation, adhesion, differentiation and tumour invasion. At the cell surface, tetraspanins form networks with a wide diversity of proteins called tetraspanin-enriched microdomains (TEMs). CD63 / Tspan-30 was the first characterized tetraspanin. In addition to its presence in TEMs, CD63 / Tspan-30 is abundantly present in late endosomes and lysosomes. CD63 / Tspan-30 is expressed in melanoma and appears to be involved in regulation of motility and adhesion of melanoma cells. CD63 / Tspan-30 is associated with early stages of melanoma tumor progression. It may play a role in growth regulation. Lack of expression of CD63 in platelets has been observed in a patient with Hermansky-Pudlak syndrome (HPS). which is a genetically heterogeneous, rare, autosomal recessive disorder characterized by oculocutaneous albinism, bleeding due to platelet storage pool deficiency, and lysosomal storage defects.
