CXADR (coxsackie virus and adenovirus receptor), also known as CAR, is a 46 kDa type I transmembrane glycoprotein expressed on the basolateral surface, and belongs to the CTX family of the Ig superfamily. It is a receptor for group B coxsackieviruses and subgroup C adenoviruses and also acts as a homophilic adhesion molecule within junctional complexes, notably between epithelial cells lining body cavities and within myocardial intercalated discs. CXADR structrually comprises two extracellular immunoglobulin domains, a transmembrane helix and a C-terminal intracellular domain. The N-terminal Ig domain (D1) of CXADR is necessary and sufficient for adenovirus binding. The C-terminus interacts with several cytoplasmic junctional proteins, microtubules and the actin cytoskeleton. Pairs of transmembrane domains of CXADR associate with each other in a detergent cloud that mimics a cellular plasma membrane. Variants of CXADR are attached to the cell membrane by a GPI- anchor.
