Beschreibung von CNTN2 / TAG-1 Protein

Contactin 2 (CNTN2), also known as TAX1 (transiently-expressed axonal glycoprotein), TAG1 (transient axonal glycoprotein), and axonin-1, is a member of the contactin subfamily belonging to the immunoglobulin superfamily. CNTN2 consists of six Ig-like domains and four fibronectin type I?II domains, and is anchored to the membrane by glycosylphosphatidylinositol (GPI), whereas the soluble form can be released by a GPI-specific phospholipase. As a neural cell adhesion molecule expressed by a subset of neuronal populations in the developing CNS and PNS, CNTN2 mediates cell-cell interactions either via homophilic, or heterophilic contacts with various adhesion molecules including NgCAM, NrCAM, NCAM and neurocan. CNTN2 thus plays an important role in cell adhesion, neurite outgrowth and axon guidance in the developing nervous system. The study data have suggested that TAG1-mediated cell-cell contact is formed by cis-assisted trans-binding. Association of TAG1 with Caspr2 (contactin-associated protein 2) is essential for the molecular organization of juxtaparanodal regions of myelinated fibers. In addition, CNTN2 is also involved in glial tumorigenesis and may provide a potential target for therapeutic intervention.
