Nachweisantikörper 0.5 mg/mL mouse anti-CDH16 monoclonal antibody conjugated to horseradish-peroxidase (HRP). Dilute to working concentration of 0.5 μg/mL in detection antibody diluteion buffer before use.

Standard Each vial contains 100 ng of recombinant CDH16. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20℃ to -70℃ in a manual defrost freezer. A seven-point standard curve usi ng 2-fold serial dilutions in sample dilution buffer, and a high standard of 7 ng/mL is recommended.

The minimum detectable dose of Human KSP-Cadherin / Cadherin-16 / CDH16 was determined to be approximately 109.4 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.

The wells are washed and a horseradish peroxidase conjugated mouse anti-KSP-Cadherin / Cadherin-16 / CDH16 monoclonal antibody is then added, producing an antibody-antigen-antibody "sandwich". The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount of KSP-Cadherin / Cadherin-16 / CDH16 present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.

Cadherins (Calcium dependent adhesion molecules) are a class of transmembrane proteins. They play important roles in cell adhesion, ensuring that cells within tissues are bound together. They are dependent on calcium(Ca2+) ions to function. Mature cadherin proteins are composed of a large N-terminal extracellular domain, a single membrane-spanning domain, and a small highly conserved C-terminal cytoplasmic domain. Type II (atypical) cadherins are defined based on their lack of an HAV cell adhesion recognition sequence specific to type I cadherins. It has been observed that cells containing a specific cadherin subtype tend to cluster together to the exclusion of other types, both in cell culture and during development.
Cadherin-16, also known as Kidney-specific cadherin, Ksp-cadherin and CDH16, is a single-pass type I membrane protein. Cadherin-16 / CDH16 is exclusively expressed in kidney, where the protein functions as the principal mediator of homotypic cellular recognition, playing a role in the morphogenic direction of tissue development. Cadherin-16 / CDH16 consists of an extracellular domain containing 6 cadherin domains, a transmembrane region and a truncated cytoplasmic domain but lacks the prosequence and tripeptide HAV adhesion recognition sequence typical of most classical cadherins.
