Mast / stem cell growth factor receptor, also known as Proto-oncogene c-Kit, Tyrosine-protein kinase Kit, CD117 and KIT, is a single-pass type I? membrane protein which belongs to the?protein kinase superfamily, Tyr protein kinase family and CSF-1/PDGF receptor subfamily. CD117 / KIT contains five?Ig-like C2-type (immunoglobulin-like) domains and one?protein kinase domain. CD117 / KIT is the receptor for stem cell factor (mast cell growth factor). It has a tyrosine-protein kinase activity. Binding of the ligands leads to the autophosphorylation of KIT and its association with substrates such as phosphatidylinositol 3-kinase (Pi3K). CD117 is the receptor for the cytokine stem cell factor (SCF), when this receptor binds to SCF it forms a dimer that activates signaling through second messengers. Signaling through CD117 plays a role in cell survival, proliferation, and differentiation. Defects in CD117 / KIT are a cause of piebaldism which is an autosomal dominant genetic developmental abnormality of pigmentation characterized by congenital patches of white skin and hair that lack melanocytes. Defects in CD117 / KIT have also been associated with testicular tumors (TEST) which is a common solid malignancy in males.
