Nacheweisantikörper 0.5 mg/mL of rabbit anti-B7-2 monoclonal antibody conjugated to horseradish-peroxidase (HRP). Dilute to working concentration of 0.4 μg/mL in detection antibody diluteion buffer before use.

Standard Each vial contains 14 ng of recombinant B7-2. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20℃ to -70℃ in a manual defrost freezer. A seven-point standard curve usi ng 2-fold serial dilutions in sample dilution buffer, and a high standard of 300 pg/mL is recommended.

The minimum detectable dose of Human CD86 / B7-2 was determined to be approximately 4.68 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.

Standards and samples are added to the wells, and any CD86 / B7-2 present binds to the immobilized antibody. The wells are washed and a horseradish peroxidase conjugated rabbit anti-CD86 / B7-2 monoclonal antibody is then added, producing an antibody-antigen-antibody "sandwich". The wells are again washed and TMB substrate solution is loaded, which produces color in proportion to the amount of CD86 / B7-2 present in the sample. To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.

The B-lymphocyte activation antigen B7-2 (referred to as B70), also known as CD86, is a member of the cell surface immunoglobulin superfamily and is constitutively expressed on interdigitating dendritic cells, Langerhans cells, peripheral blood dendritic cells, memory B cells, and germinal center B cells. B7-2 exists predominantly as a monomer on cell surfaces and interacts with two co-stimulatory receptors CD28 and cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) expressed on T cells, and thus induces the signal pathways which regulate T cell activation and tolerance, cytokine production, and the generation of CTL. Binding of B7-2 with CD28 induces the activation of T cell, whereas CTLA-4 acts as a negative regulator and diminishes the immune responses. Recent study has revealed that B7-2 promotes the generation of a mature APC repertoire and promotes APC function and survival. It is indicated that contacts between B and T helper cells mediated by CD86 encourage signals for the proliferation and IgG secretion of normal B cells and B cell lymphomas. Furthermore, the B7 proteins are also involved in innate immune responses by activating NF-κB-signaling pathway in macrophages. CD86 is regarded as a promising candidate for immune therapy.
