Preparation

This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Ferret CD4 (rF CD4;Catalog#60003-F08H; ABS50090.1; Met 1-Leu 401). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.

Western blot This antibody can be used at 1-2 μg/mL with the appropriate secondary reagents to detect Ferret CD4 in WB. Using a DAB detection system, the detection limit for Ferret CD4 is approximately 2 ng/lane under non-reducing conditions and 1 ng/lane under reducing conditions.
Direct ELISA This antibody can be used at 0.5-1 μg/mL with the appropriate secondary reagents to detect Ferret CD4. The detection limit for Ferret CD4 is approximately 0.078 ng/well.
Storage This antibody can be stored at 2℃-8℃ for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -70℃. Preservative-Free.
Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

Hintergrund von CD4 Antikörper

T-cell surface glycoprotein CD4, also known as T-cell surface antigen T4/Leu-3, is a single-pass type I membrane protein. CD4 contains three Ig-like C2-type (immunoglobulin-like) domains and one Ig-like V-type (immunoglobulin-like) domain. CD4 is a glycoproteinexpressed on the surface of T helper cells, regulatory T cells,monocytes, macrophages, anddendritic cells. It was discovered in the late 1970s and was originally known as leu-3 and T4 before being named CD4 in 1984. CD4 is aco-receptorthat assists theT cell receptor (TCR) to activate its T cell following an interaction with anantigen presenting cell. Using its portion that resides inside the T cell, CD4 amplifies the signal generated by the TCR. CD4 interacts directly withMHC class II molecules on the surface of the antigen presenting cell via itsextracellulardomain. CD4 is a primary receptor used byHIV-1to gain entry into host T cells. HIV infection leads to a progressive reduction of the number of T cells possessing CD4 receptors. Therefore, medical professionals refer to the CD4 count to decide when to begin treatment for HIV-infected patients.
