Standard Each vial contains 235 ng of recombinant CA14. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20℃ to -70℃ in a manual defrost freezer. A seven-point standard curve usi ng 2-fold serial dilutions in sample dilution buffer, and a high standard of 8 ng/mL is recommended.

The minimum detectable dose of Human Carbonic Anhydrase XIV / CA14 was determined to be approximately 125 pg/ml. This is defined as at least three times standard deviations above the mean optical density of 10 replicates of the zero standard.

To end the enzyme reaction, the stop solution is added and absorbances of the microwell are read at 450 nm.

CA XIV is a polypeptide consisting of an extracellular N-terminal catalytic domain, a membrane-spanning segment and a short intracellular C-terminal segment with several potential phosphorylation sites. The CA XIV protein is expressed in the brain on plasma membranes of neuronal bodies and axons, in the kidney preferentially on the apical membranes of proximal tubules, and in the liver on the plasma membranes of hepatocytes. CA14, together with CA4, play important roles in the regulation of intracellular pH in hippocampal neurons by facilitating AE3-mediated Cl––HCO3- exchange.
