The recombinant heterotrimer of human AMPK (PRKAG1 / PRKAB2 / PRKAA1) has a calculated molecular mass of 160 (38+30+92) KDa. The apparent molecular mass is approximately 35, 37 & 95 KDa respectively in SDS-PAGE under reducing conditions.

Formulation Lyophilized from a 0.2μm filtered solution of 50mM Tris ,200mM NaCl,2mM GSH,1mM EDTA ,1mM DTT,10%gly, pH 7.4.

AMP-activated protein kinase (AMPK) is an important metabolic stress-sensing protein kinase induced by AMP. It can be detected in a number of tissues, including the liver, brain, and skeletal muscle. AMPK is a heterotrimer comprised by three proteins (α, β, and γ subunits) that together make a functional enzyme, conserved from yeast to humans. Seven known genes encode the subunit isoforms (α1, α2, β1, β2, γ1, γ2, γ3) and all are expressed in the heart. The most common isoforms expressed in most cells are the α1(PRKAA1), β1(PRKAB1), and γ1(PRKAG1) isoforms. AMPK induces a cascade of events within cells in response to the ever changing energy charge of the cell. Mammalian AMPK is a stable alphabetagamma heterotrimer comprising a catalytic alpha (PRKAA) and two non-catalytic subunits, beta (PRKAB) and gamma (PRKAG). AMPK activation results in the stimulation of hepatic fatty acid oxidation and ketogenesis, inhibition of cholesterol synthesis, lipogenesis, and triglyceride synthesis, inhibition of adipocyte lipolysis and lipogenesis, stimulation of skeletal muscle fatty acid oxidation and muscle glucose uptake, and modulation of insulin secretion by pancreatic beta-cells.
