Usage Guide von ALK-3 / BMPRIA Antikörper

Specificity

Mouse ALK-3 / BMPRIA

Western blot This antibody can be used at 1-2 μg/mL with the appropriate secondary reagents to detect Mouse ALK3 in WB. Using a DAB detection system, the detection limit for Mouse ALK3 is approximately 0.5 ng/lane under non-reducing conditions and 0.25 ng/lane reducing conditions.
Direct ELISA This antibody can be used at 0.1-0.2 μg/mL with the appropriate secondary reagents to detect Mouse ALK3. The detection limit for Mouse ALK3 is approximately 0.00245 ng/well.
Storage This antibody can be stored at 2℃-8℃ for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -70℃.Preservative-Free.
Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

Hintergrund von ALK-3 / BMPRIA Antikörper

ALK3, also known as BMPR1A, is a type I receptor for bone morphogenetic proteins (BMPs) which belong to the transforming growth factor beta (TGF-β) superfamily. The BMP receptors form a subfamily of transmembrane serine/threonine kinases including the type I receptors BMPR1A and BMPR1B and the type II receptor BMPR2. Type I and II BMP receptors are both indispensable for signal transduction. Upon ligand binding, they form a heterotetrameric-activated receptor complex consisting of two pairs of a type I and a II receptor. Type II receptors can bind ligands in the absence of type I receptors, whereas the respective type I receptors are required for signal transduction. The downstream molecules of the type I BMP receptors include the Smad (Smad1, 5 and 8) proteins that are phosphorylated in a ligand-dependent manner, and relay the BMP signal from the receptors to target genes in the nucleus. The expression of BMP type I receptors is up-regulated in bone morphogenesis and play central roles during embryonic development and fracture healing. In addition, it has been shown that type 1B and 1A BMP receptors transmit different signals to bone-derived mesenchymal progenitors and function in both the specification and differentiation of osteoblasts and adipocytes. Germline BMPR1A mutations cause a significant proportion of cases of Juvenile polyposis syndrome (JPS).
